Method summary
The original move of wtPAR-2 (tiff format) is rotated at 45 degree and processed through Rolling Ball and Kalman filters to obtain wtPAR-2_RBKL.avi (Movie S1). The PAR-2 trajectories were obtained from the avi file. Kalman filter slightly reduced the diffusion coefficient.
