3D images of mitochondria in living HeLa cells with conventional SPA-SIM with single-photon light sheet activation

3D images of mitochondria visualized by Skylan-NS in living HeLa cells with selective-plane-activation structured illumination microscopy (SPA-SIM) using single-photon light sheet activation were obtained. SPA-SIM was used to visualize the mitochondrial components distributed inside the cells owing to out-of-focus rejection. Skylan-NS is a photoswitchable green fluorescent protein. CW laser is used for single-photon activation, which is more efficient and allows measurement with higher temporal resolution.
