Fluorescence images of spheroid of living HeLa cells labeled by rsGamillus-S in SPA3DSIM mode for two-photon activation

Fluorescence images of histone H2B in the cell spheroids of living HeLa cells labeled by rsGamillus-S were obtained in SPA-3DSIM mode using two-photon activation. SPA-3DSIM achieved super-resolution imaging of histone H2B distribution in a spheroid. rsGamillus-S is an acid-resistance photoswitchable fluorescent protein, which owns exceptionally high on-switching rate to show the nuclei more clearly. Two-photon activation using a scanned Bessel beam produces a wider FOV and a higher axial resolution.
