Fluorescence images of spheroid of living HeLa cells labeled by rsGamillus-S in 3DSIM mode for two-photon activation

Fluorescence images of histone H2B in the cell spheroids of living HeLa cells labeled by rsGamillus-S were obtained in three-dimensional structured illumination microscopy (3DSIM) mode using two-photon activation. The 3DSIM mode was difficult for imaging the internal structure of spheroid. rsGamillus-S is an acid-resistance photoswitchable fluorescent protein, which owns exceptionally high on-switching rate to show the nuclei more clearly. Two-photon activation using a scanned Bessel beam produces a wider FOV and a higher axial resolution.
