Fluorescence images of histone H2B in the nucleus of living HeLa cells labeled by rsGamillus-S were obtained in selective-plane-activation structured illumination microscopy (SPA-SIM) mode using two-photon activation. SPA-SIM achieved the observation of the internal structures of the nucleus with high spatial resolution and image contrast. rsGamillus-S is an acid-resistance photoswitchable fluorescent protein, which owns exceptionally high on-switching rate to show the nuclei more clearly. Two-photon activation using a scanned Bessel beam produces a wider FOV and a higher axial resolution.
