Description
Fluorescence images of histone H2B in the nucleus of living HeLa cells labeled by rsGamillus-S were obtained in structured illumination microscopy (SIM) mode using two-photon activation. SIM improved the spatial resolution and optical sectioning capability. Internal structures were buried in the background light. rsGamillus-S is an acid-resistance photoswitchable fluorescent protein, which owns exceptionally high on-switching rate to show the nuclei more clearly. Two-photon activation using a scanned Bessel beam produces a wider FOV and a higher axial resolution.
