Fluorescence images of histone H2B in the nucleus of living HeLa cells labeled by rsGamillus-S in 3DSIM mode for two-photon activation

Fluorescence images of histone H2B in the nucleus of living HeLa cells labeled by rsGamillus-S were obtained in three-dimensional structured illumination microscopy (3DSIM) mode using two-photon activation. 3DSIM improved the spatial resolution and optical sectioning capability. Internal structures were buried in the background light. rsGamillus-S is an acid-resistance photoswitchable fluorescent protein, which owns exceptionally high on-switching rate to show the nuclei more clearly. Two-photon activation using a scanned Bessel beam produces a wider FOV and a higher axial resolution.
