Fluorescence images of mitochondria in fixed HeLa cells in the imaging mode of SPA3DSIM
Description
The fluorescence images of mitochondria labeled with Skylan-NS in fixed HeLa cells were taken and reconstructed in the imaging mode of SPA-3DSIM. The optical sectioning capability in SPA-3DSIM mode had further improvements. The out-of-focus light in the image produced by the SPA-3DSIM was reduced more efficiently, while the practical spatial resolution was slightly inferior. Skylan-NS is a photoswitchable green fluorescent protein. CW laser is used for single-photon activation, which is more efficient and allows measurement with higher temporal resolution.
