Title
3D medaka brain visualized clonally related units in the pallium
Description
The dorsal telencephalon (i.e. the pallium) exhibits high anatomical diversity across vertebrate classes. The non-mammalian dorsal pallium accommodates various compartmentalized structures among species. The developmental, functional, and evolutional diversity of the dorsal pallium remain unillustrated. Here, the authors analyzed the structure and epigenetic landscapes of cell lineages in the telencephalon of medaka fish (Oryzias latipes) that possesses a clearly delineated dorsal pallium (Dd2). They found that pallial anatomical regions, including Dd2, are formed by mutually exclusive clonal units, and that each pallium compartment exhibits a distinct epigenetic landscape.

Z-stack images of horizontal section of medaka telencephalon visualizing units of GFP-positive cells, each of which has the same lineage
Description
Z-series images of medaka brain obtained by light sheet microscopy that visualize clonally related units in the pallium. This transgenic line express fluorescent protein under HuC promoter, which drives expression in neural progenitor cells. As the reporter GFP was expressed in neural progenitor cells at the neurula stage (stage 16–17) by Cre-loxp recombination system, we can trace the lineage in the adult fish. We can see some GFP-positive clonally related compartments that distribute exclusively in one anatomical region but not across many anatomical regions. Description
The medaka brain deriverd from two transgenic lines. One is the transgenic line (Tg (HuC:loxp-DsRed-loxp-GFP)) that visualized neural progenitors by the HuC promoter. The second is a line (Tg (HSP-Cre)) that expresses Cre recombinase under the heat shock protein promoter. In the absence of the recombination, DsRed is expressed in the neural progenitor cells. When Cre-loxp recombination is induced the neural progenitors express GFP instead of DsRed.
